There are four known species of Plasmodium which can infect human being, namely P. falciparum Rvivax, P.malariae, and Rovale. They differ in mode of transmission, clinical manifestation, response to drug and immune response of the infected person. All these differences indicate that there is a species - specificity of the plasmodium parasite. Therefor, it is very important to identify the species of the parasite which cause malaria in an endemic area.The microscopic examination of Giemsa stained thick blood smear is often the only diagnostic method available in the laboratory in Indonesia. Only well trained and experienced morphologist can make a correct identification of the infecting species. In this study semi-nested PCR technic was applied for the identification of the infecting parasite.This technic is sensitive and specific. Thirty five blood samples of infected persons from Trenggalek Regency, Fast Java were examined. We have found 48% persons were infected by P. falciparum 4 % by P, vivax 16 % mixed infection by P, falciparum and P.vivax, 20% mixed infection by Pfalciparum and P.malariae, 4% mixed infection by P.vivax and P.malariae, and 8% mixed infection by P. falciparum Rvivax, and P.malariae. There was no single infection by Rmalariae nor Rovale. This finding was interesting because P. malariae was never been reported before in Trenggalek area.
