Elevated esterase activity is one of the mechanisms of resistance to organophosphate insecticides in mosquitoes Such physiological resistance can be detected by a microplate assay method. Culex quinquefasciatus Say is one of the domestic mosquitoes of medical importance showing risk of exposure to organophosphate insecticides applied indoor either as thermal fog or ultra low volume (ULV) since the last two decades in urban areas in Indonesia including Yogyakarta. The present study aims at determining the potency of Cx. Quinquefasciatus breeding in Yogyakarta to be resistant to insecticiades related to esterase activity mechanism. A microplate assay with a-naphtvl acetate as the substrate and Fast Blue B as the coupling reagent and homogenates of late third or early fourth instars of the larval mosquito were used during the study. It was concluded that Cx. quinquefasciatus larvae collected from some districts in Yogyakarta was mostly susceptible and showed a low degree of potency of being resistant to the esterase-related insecticides.
