Study on prevalence of glucose 6-phosphate dehydrogenase (G6PD) deficiency has been carried out in populations of two malaria endemic regions in Aceh Province. A total number of 138 individuals consisted of 50 Great Acehnese (32 male and 18 female) and 89 Middle Acehnese (56 males and 33 female) were involved in the study. Blood of all participants were collected in EDTA-tubes and screened for detection of G6PD deficiency using .fluorescence spot test. The G6PD deficiency was found in 2 (4.0%) out of 50 Great Acelmese and in 1 (1.12%) out of 89 Middle acehnese No female subject was G6PD deficient, but four out of 12 female Great acehnese were detected having G6PD variant with intermediary activity. From the results and some additional reports of malarial cases in the areas, it is assumed that endemicity status of Great Aceh Municipality is higher than that of Middle Aceh Municipality.
