Abstract

Immunomodulation effects of aquaous and methanol extracts of kencur (Kaempferia galanga Linn) were studied for phagocytic effect in vitro.Methanol extract of kencur was also studied for phagocytic effect in vivo. It is known that methanol extract of keneur has stronger suppressive effect than aquaous one. This is probably due to p-methoxy einnamat ethyl ester contained within methanol extracts of kencur. The aim of this study was to investigate the immunosuppressive effect of p-methoxy cinnamat ethyl ester in kencur on phagocytosis in vitro. For this purpose an in vitro granulocyte phagocytosis assay was used (Wagner, et al. 1991). This method assesses the speed of neuthrophyl ingestion on antigen after treated with test substance.The source of neutrophil was taken from five healthy adults and then separated from other cells using ficoll They were divided into experimental and control group. Experimental group was further divided into three subgroups receiving of 100 ug/ml 150 uglml and 200 ug/ml p-methoxy cinnamat ethyl ester. Control group received the solvent compound of p-methoxy cinnamat ethyl ester. Phagocytic effect can be seen from the incubating result of nuethrophyl with yeast cells (Sacchromyces cereviceae), to enable us to count phagocytosis index (PI) and the phago percentages (PP). The PP of neuthrophyls treated with p-methoxy cinnamat ethyl ester were - 7.826% of 100 (g/ml and -19.909% for 200 (glml concentration respectively. The negative value showed that p-methoxy cinnamat ethyl ester compound has immunosuppressive effect on the phagocytosis capability of neuthropyl in vitro.
