Some clinical investigators have noted that estrogen may improve cognitive function. Various studies suggest several mechanisms by which estrogen may affect neuropsychological functions, namely modulation of neurotransmitters, neurit and synaptic reorganization, prevention of cerebral ischemia and lipoprotein alterations. The objective of this study is to reveal the effect of dose and duration of estrogen administration on spatial memory of the rat by measuring the performance in the eight-arm radial maze.In the present study, 3 month female rats (n= 40) were devided randomly into 8 groups, i.e. 4 treatment groups (P1, P2, P3 and P4) and 4 control groups (KP1, KP2, KP3 and KP4). Rats in the treatment groups were ovarectomized and given 0,01 mg/kg bodyweight (PI) and 0,005 mg/kg bodyweight (P2) etinyl estradiol, orally, in two weeks; and given 0,01 mg/kg bodyweight (P3) and 0,005 mg/kg bodyweight (P4) etinyl estradiol orally, in four weeks. Rats in the control groups were ovarectomized and given olive oil solution, orally, in two weeks (KP1 and KP2) or four weeks (KP3 and KP4). Within 12 days, a test of eight-arm radial maze was conducted for individual rat to observe its performance.The results of this study showed that the performance as assessed by revealing correct choice accuracy and error type A were better in the treatment groups than control groups. Correct choice accuracy (%) were 94,81 (P1), 93,60 (P2), 95,24 (P3), 98,15 (P4), 18,13 (KP1 37,99 (KP2), 55,56 (KP3) and 48,.91 (KP4); error type A (%) were 5,18(P1), 6,39 (P2),4,76 (P3), 1,85 (P4), 37,19 (KP1), 23,91 (KP2), 19,26 (KP3) and 39,85 (KP4).From the results of the present study it is concluded that estrogen could enhance spatial memory, depend on dose and duration of estrogen administration, as shown by the performance of the traetment groups which is better than the control groups
